From his profile I wouldn't have guessed John Wayne, but his voice is recognizable.

If I had seen him for the first time in The Big Trail, I would not have expected him to grow up into "John Wayne." I'm not even sure I would have expected him to specialize in Westerns. I would have figured him for a heck of a lot more romantic parts. Dialogue you can teach, chemistry happens.
